If it is a blood test (vs breath) there are two basic routes for testing. In both instances, the blood would be drawn from the antecubital vein in the arm. This is usually done by a nurse, physician or phlebotomist at a hospital. Most hospitals (depends on the state) measure alcohol in serum (the watery part of the blood) or in actual whole blood. If you were arrested for some crime (e.g., DUI), police may request a blood sample be drawn at the hospital then it is transferred to a forensic lab where, in most cases, blood is tested for alcohol. Most hospitals use a different analytical method than that used by forensic labs but both results are accurate when correctly interpreted. The actual analysis takes about 5 minutes if everything is set up and ready to go, but the normal turn around time varies on need. In a hospital setting, such results are usually available within a hour or two (when ordered "STAT"). That's because patient diagnosis treatment issues could be affected by alcohol so docs need to know. In a forensic lab, it often takes about 1-2 weeks just because of the volume of work and the need to often do duplicate analyses. If you are a high profile person, the lab could probably turn the results around in a few hours if pressured to do so by the DA or Prosecutor.
